Access to high-quality plant genome assemblies has greatly advanced our understanding of species diversity. The significant reduction in Long Read sequencing costs has democratized access to biodiversity studies. However, exploring intra-species variability within regions of interest (ROI), especially in large genomes, remains expensive. In this context, targeted enrichment methods emerge as a strategic solution for analyzing biodiversity across broader sample sets. These approaches provide precise and reliable data, enabling the identification of genomic regions associated with key traits in specific genotypes.


The French Genomic Center (CNRGV) provides three cutting-edge/innovative strategies for efficiently capturing large regions of interest in complex plant genomes, including Twist Target Enrichment. To demonstrate the potential of this approach, we conducted a pilot project to characterize a 60Kb locus involved in pathogen resistance in common bean. We then further validated the technology by applying it to a more complex plant species, bread wheat, to characterize a 750Kb region of interest.
