How do I onboard my Custom Vector?

To onboard your custom vector, please follow these steps:

  • Download and complete the Custom Vector submission form. 
  • Email the form to customer support: customersupport@twistbioscience.com. 
  • Send an aliquot (see below for details) of the custom vector DNA to:

  • Twist charges a one-time onboarding fee of $500 per custom vector and $250-500 per additional insertion site.
  • Based on the availability of the queue, your vector will be assigned an onboarding start date once both of your form and vector have been received.  
  • Onboardings start every Monday. It will take 10-15 business days for your vector to complete the onboarding process from the start date.
  • Once vector onboarding is complete, you will be able to select it when ordering Clonal Genes from our online website. Please note that we cannot start synthesis of inserts during the onboarding process. Additionally, do not use the Custom Vector submission form to submit your gene orders - the insert sequence in your vector onboarding form will not be synthesized as part of the onboarding process. 

Please ensure your vector meets the following requirements:

  • Vector Volume ≥ 100 uL (Plasmid material cannot be submitted on filter paper)
  • Vector Concentration ≥ 100 ng/uL
  • Vector Mass 10 ug
  • Must have bacterial origin of replication
  • Must be clonal population
  • Must be circular
  • Must have antibiotic resistance marker
  • Must not contain homopolymers/repetitive sequence >9bp within 40bp of the insertion site
  • Repeats: Must contain at least 40bp of unique sequence with no direct or inverted repeats >19bp upstream and downstream of the insertion site
  • Must not contain ccdB in final gene product
  • Must be able to linearize by Restriction Enzyme site within 50bp of insertion site
  • If your vector requires a Stbl-like cell line, it cannot contain ccdB in initial construct material sent to Twist
  • Must be BSL-1 compliant

Antibiotic Resistance:

  • Plasmid must contain one of the below antibiotic resistance markers for E.coli.
  • Standard concentrations are below:
