Twist Biopharma has developed a set of antibodies that targets the SARS-CoV-2 Spike
Protein with high binding affinity and specificity. Several antibodies also show the ability to
block S1 binding to ACE2.
We have also developed a set of antibodies targeting the extracellular domain of the ACE2
receptor. These antibodies have high binding affinity and have been demonstrated to inhibit
the binding of the S1 protein to ACE2 in vitro.
All antibodies are fully human and are either hIgG1, IgG2, or IgG1 VHH Fc making them ideal
as potential candidates for both diagnostic and therapeutic uses.

SARS-CoV-2 スパイクタンパク質を標的とした Twist Biopharma のヒト抗体は、治療におけるウイルス中和と、診断におけるウイルス検出の２つの目的で用いることが可能です。
こちらは、 SARS-CoV-2 スパイク糖タンパク質 S1 または S1 三量体タンパク質に結合する各抗体クローンの親和性データです。各クローンは異なる色で表されています（凡例参照）。
抗 S1 抗体に、ACE2 受容体を発現する細胞への標識 S1 タンパク質の結合を阻害する能力があるかどうか明らかにするために、細胞ベースの S1ーACE2 競合アッセイを行いました。下図に示すように、いくつかの抗体が、標識 S1 RBD が Vero 細胞上の ACE2 に結合するのを阻害しています（TB182-03、TB182-04、TB182-06、TB182-13、TB181-63）。

抗ヒト ACE2 受容体抗体は、ACE2 受容体に対して高い親和性を示すことから、治療用の遮断抗体または競合抗体として有望と考えられます。
抗 ACE2 抗体は、ピコモルからナノモルの範囲の親和性を持ち、抗原標的に対して非常に高い特異性と親和性を示します。このアッセイで得られた Fc キャプチャおよびダイレクトキャプチャ（DC）データを下表に示します。
パネル全体としては、下表に示すように、VERO E6 細胞に対する特異的な結合が幅広く見られます。単一クローンのフローサイトメトリーデータも例として示しています。ACE2 結合のポジティブコントロールとして、S1 Fc 融合タンパク質および S1 RBD Fc 融合タンパク質（自社発現）についてのデータも示しています。
S1 タンパク質の ACE2 への結合を妨げる各抗体クローンの能力を評価するために、ACE2 の段階希釈液をビオチン化 ACE2 タンパク質に添加しました。この混合物を ELISA プレートに固定化した SARS-CoV-2 S タンパク質 RBD に添加し、ストレプトアビジン-HRP で検出しました。対数変換とカーブフィッティングは、データポイントからバックグラウンドを除いてから行いました。すべての抗体クローンについて、IC50 値を表に示します。
