Libraries converted for methylation sequencing pose unique challenges to target enrichment because they are less complex than typical libraries. To overcome these obstacles, Twist optimized the Twist NGS Methylation Detection System with two key advancements: adaptive panel design and a methylation-specific blocker called Methylation Enhancer. This technical note outlines how these optimizations provide both flexibility and unprecedented control over the off-target rate in targeted methylation sequencing.
