Targeted sequencing, whole exome sequencing in particular, effectively focuses sequencing costs on genomic regions of interest, and for the parallel and simultaneous interrogation of thousands of targets, hybridization-based target enrichment methods are preferred. These methods, however, do not always fulfill the promise of cost-efficiency. Context-specific biases and non-uniformity, particularly in highly repetitive, GC-rich and non-unique regions of the genome, arise as a consequence of design and synthesis limitations and suboptimal workflow stringency, and can reduce overall data quality and assay efficiency. Twist Bioscience has entered the targeted sequencing market with novel Target Enrichment Solutions that liberate researchers from these limitations. Twist’s scalable solutions combine quick customization and content flexibility with unprecedented sequencing efficiency. Double-stranded DNA probes can be individually tuned to enrich target regions with exceptional uniformity, reducing the overall cost of sequencing. Twist Bioscience’s fast and precise DNA Synthesis Platform also allows iterative testing of custom panels before locking in the final design - all while still providing exceptional uniformity. Continuing to innovate, Twist now offers a faster solution to hybridization that allows researchers to move from sample to sequencer in a single day. Additional applications for detection of cancer biomarkers have been developed with FFPE and differentially methylation regions. This presentation presents data that illustrate target enrichment efficiency through diverse sample qualities using Twist’s complete solution. The data will demonstrate how Twist enables researchers to sequence more deeply and more efficiently into each sample.
