Automated high-throughput DNA synthesis and assembly
Abstract
DNA synthesis and assembly primarily revolve around the innovation and 3 refinement of tools that facilitate the creation of specific genes and the manipulation 4 of entire genomes. This multifaceted process encompasses two fundamental steps: the 5 synthesis of lengthy oligonucleotides and the seamless assembly of numerous DNA 6 fragments. With the advent of automated pipetting workstations and integrated 7 experimental equipment, a substantial portion of repetitive tasks in the field of 8 synthetic biology can now be efficiently accomplished through integrated liquid 9 handling workstations. This not only reduces the need for manual labor but also 10 enhances overall efficiency. This review explores the ongoing advancements in the 11 oligonucleotide synthesis platform, automated DNA assembly techniques, and 12 biofoundries. The development of accurate and high-throughput DNA synthesis and 13 assembly technologies presents both challenges and opportunities.
