We can now explore sequence space in a targeted, deterministic fashion to build a holistic understanding of the mechanisms of protein binding. Regulatory elements have been studied deeply with traditional 120 base pair libraries, but in reality many regulatory elements are much longer than that. Using 450 base pair Multiplexed Gene Fragments from Twist we are now able to systematically screen for regulatory elements more finely tuned to drive gene expression in our specific cell type.
