One of the primary applications of genomics is to connect genotype to phenotype. Linkage mapping utilizes recombination to identify regions of the genome that code for traits of interest. Our lab has used the cost-effective high-throughput Twist 96-Plex Library Prep (Riptide) kit in multiple crosses to perform whole-genome skim sequencing and create high-quality genetic maps. These genetic maps have been used to identify regions of the genome and, using quantitative trait locus (QTL) analysis, specific candidate genes responsible for various plant traits. I will demonstrate our process from raw genomic DNA to a genetic map and QTL. This process can be applied to and adapted for any diploid system.
